Meta-analysis of three genome-wide association studies identifies two loci that predict survival and treatment outcome in breast cancer

Abstract: The majority of breast cancers are driven by the female hormone oestrogen via oestrogen receptor (ER) alpha. ER-positive patients are commonly treated with adjuvant endocrine therapy, however, resistance is a common occurrence and aside from ER-status, no unequivocal predictive biomarkers are currently in clinical use. In this study, we aimed to identify constitutional genetic variants influencing breast cancer survival among ER-positive patients and specifically, among endocrine-treated patients. We conducted… Show more

“…Although experimental data suggests that VEGFA and IL-1β play a role in BC, our findings, along with the results from several other studies (22)(23)(24), do not suggest that several common polymorphisms in these genes are associated with BC clinical and morphological variables and BC survival rates.…”

“…The authors, however, conducted no DFS, MFS or OS analyses. Furthermore, none of the GWAS studies showed a statistically significant association between IL-6 polymorphisms and BC survival (23,24). The contrasting findings in IL-6 polymorphism studies may be due to sampling errors or differences in patient ethnicity.…”

“…We also found protective associations between higher GReX of AURKA (20q13.2) , PIK3CA (3q26.32) , SERPINB5 ( 18q21.33 ) and lower risk of breast cancer-mortality (Figure 4C). Of these 4 loci, associations with survival have been reported with SNPs in the same chromosomal region as AURKA , PIK3CA, and SERPINB5 [8,29–33], though none of these reported SNPs were utilized in constructing the GReX of this gene. Furthermore, the GReX of these four genes were not significantly correlated ( P > 0.05 for all pairwise Spearman correlation tests), and the sets of SNPs used in constructing the GReX of these four genes had no pairwise intersections, providing evidence that their independent association with breast cancer-specific survival was not a pleiotropic effect from shared or correlated SNPs.…”

“…Haplotype block refers to the inheritance of a cluster of SNPs [28]. LDlink (https://analysistools.nci.nih.gov/LDlink/) was used to calculate pair-wise linkage disequilibrium (LD) among SNPs associated with the same gene.…”
