Research Review: Brain network connectivity and the heterogeneity of depression in adolescence – a precision mental health perspective

Abstract: Background Adolescence is a period of high risk for the onset of depression, characterized by variability in symptoms, severity, and course. During adolescence, the neurocircuitry implicated in depression continues to mature, suggesting that it is an important period for intervention. Reflecting the recent emergence of ‘precision mental health’ – a person‐centered approach to identifying, preventing, and treating psychopathology – researchers have begun to document associations between heterogeneity in feature… Show more

“…In this context, researchers are beginning to use neuroimaging to identify biomarkers of risk and resilience related to the onset and course of psychopathology ( 30 ). Emerging research suggests that the intrinsic functional architecture of brain networks (i.e., ensembles of brain regions that co-activate when a person is at “rest,” or not engaged in a task) may reflect differential adaptation to stress ( 31 ), particularly in individuals at risk for psychopathology ( 32 ).…”

“…Major depressive disorder (MDD) is a heterogeneous syndrome varying considerably in clinical presentation, progression, treatment response and neurobiology. 1 Failure to effectively resolve this heterogeneity, especially in biological terms, limits drug discovery and personalised treatment approaches for patients with MDD. Most data-driven MDD subtyping efforts have focused on discriminating different symptom profiles.…”

“…within-and between-network RSFC) may provide insight into affective processes that are otherwise masked by the use of group-averaged approaches (e.g. comparing participants with a disorder to healthy volunteers) such as in the case of adolescent depression (Chahal, Gotlib, & Guyer, 2020) or by examining relations with a specific symptom or set of symptoms. For example, adults with depression have been differentiated from healthy adults based on the connectivity-based subgroup (i.e.…”
