ZFP36L2 is required for self-renewal of early burst-forming unit erythroid progenitors

Abstract: Stem cells and progenitors in many lineages undergo self- renewing divisions, but the extracellular and intracellular proteins that regulate this process are largely unknown. Glucocorticoids stimulate red cell formation by promoting self-renewal of early erythroid burst forming unit-erythrocyte (BFU-E) progenitors1-4. Here we show that the RNA binding protein Zfp36l2 is a transcriptional target of the glucocorticoid receptor (GR) in BFU-Es and is required for BFU-E self-renewal. Zfp36l2 is normally downregulat… Show more

“…However, the functional significance of these motifs has remained unknown. Two paralogs of TTP exist in the human genome, BRF1 (also known as ZFP36L1 and Tis11b) and BRF2 (ZFP36L2, Tis11d), which, like TTP, promote degradation of ARE-containing mRNAs (Ciais et al 2004;Hodson et al 2010;Zhang et al 2013). These proteins share the conserved zinc finger and CIM domains with TTP, but lack the tetraproline motifs characteristic of TTP.…”

“…[96][97][98][99] Similarly, stem cell factor (Scf) and its receptor, c-Kit, have an important role in the expansion of stress erythroid progenitors via Erk and Akt, which is enhanced by the concomitant activation of the glucocorticoid receptor (GR). 100 Additional factors that might contribute to SE are the RNA-binding protein ZFP36L2, 101 the Notch receptor 2, 102 the anti-inflammatory polymeric immunoglobulin A1 (pIgA1) 103 and dexamethasone. 104 In particular, dexamethasone, in addition to inducing proliferation of proerythroblasts, 105 stimulates expansion of these cells indirectly by supporting the activity of macrophages.…”

“…The earliest committed erythroid progenitor is the burstforming unit erythroid (BFU-E) (12,13). BFU-Es undergo limited self-renewal divisions and differentiate to the colonyforming unit erythroid (CFU-E) (14). Morphologically distinguishable stages of maturation compose the process of terminal erythropoiesis that begins at the CFU-E stage and involves three to six terminal divisions (depending on the species and developmental stage); induction of erythroid-important genes, chromatin condensation; and, in mammals, enucleation.…”
